EUR 638.4
Description: Description of target: This gene encodes a cytokine that functions in inflammation and the maturation of B cells. In addition, the encoded protein has been shown to be an endogenous pyrogen capable of inducing fever in people with autoimmune diseases or infections. The protein is primarily produced at sites of acute and chronic inflammation, where it is secreted into the serum and induces a transcriptional inflammatory response through interleukin 6 receptor, alpha. The functioning of this gene is implicated in a wide variety of inflammation-associated disease states, including suspectibility to diabetes mellitus and systemic juvenile rheumatoid arthritis. Alternative splicing results in multiple transcript variants.;Species reactivity: Human;Application: ;Assay info: Assay Methodology: Quantitative Sandwich ELISpot Immunoassay;Sensitivity:

PARPtrap™ Assay Kit for PARP1

80584-1 96 rxns.
EUR 2145
Description: The PARPtrap™ Assay Kit for PARP1 is designed to measure PARP1/DNA complex formation in a high throughput screening assay using fluorescence polarization (FP). PARP1 is known to bind damaged DNA through its DNA-binding domains. Binding to DNA activates PARP1 and in the presence of NAD+ PARP1 ribosylates itself (auto-ribosylation), what in consequence leads to PARP1 dissociation from the DNA due to the accumulated negative charge of the ribosyl polymer. In the presence of some inhibitors, however, PARP remains bound to the DNA, a phenomenon termed trapping. Trapped PARP-DNA complexes have been shown to be highly cytotoxic to cancer cells, therefore such inhibitors may be desirable for cancer treatment. The key to the PARPtrap™ Assay Kit for PARP1 is the fluorescent-labeled oligonucleotide duplex. In the absence of ribosylation, PARP1 binds to the fluorescent probe, forming a large complex and resulting in the emission of highly polarized light. However, after auto-ribosylation, PARP1 dissociates from the oligonucleotide duplex, which then rotates freely, emitting less polarized light (Fig. 1). Addition of a PARP1 inhibitor results in trapping of PARP1 to the fluorescent oligonucleotide duplex, and increases the FP signal in a dose dependent manner. The PARPtrap™ Assay Kit for PARP1 is a fluorescence polarization homogeneous assay. The FP signal is measured using a fluorescent microplate reader capable of measuring fluorescence polarization.

PARPtrap™ Assay Kit for PARP2

EUR 1950
Description: The PARPtrap™ Assay Kit for PARP2 is designed to measure PARP2/DNA complex formation in a high throughput screening assay using fluorescence polarization (FP). Similarly, to PARP1, PARP2 recognizes and binds damaged DNA via its DNA-binding domain. Binding to DNA activates PARP2 and in the presence of NAD+ PARP2 ribosylates itself (auto-ribosylation), which leads to PARP2 dissociation from the DNA due to the accumulated negative charge of the ribosyl polymer. Sirtuin 6 Fluorogenic Assay Kit

50022 96 rxns.
EUR 445
Description: The Sirtuin 6 Fluorogenic Assay Kit is a complete assay system designed to measure SIRT6 activity for screening and profiling applications. It comes in a convenient 96-well format, with all the reagents necessary for 100 fluorescent SIRT6 activity measurements. In addition, the kit includes purified SIRT6 enzyme and a SIRT inhibitor, Nicotinamide, for use as a positive and negative control, respectively. The Fluorogenic SIRT6 Assay Kit is based on a unique fluorogenic substrate and developer combination. This assay method eliminates dealing with the radioactivity, extraction, and chromatography aspects of traditional assays. Using this kit, only two simple steps on a microtiter plate are needed to analyze the SIRT6 activity level. First, the Sirtuin 6 fluorometric substrate, containing a myristoylated lysine side chain, is incubated with purified SIRT6 enzyme. The demyristoylation sensitizes the substrate so subsequent treatment with the SIRT Developer produces a fluorophore that can then be measured using a fluorescence reader.

EUR 385
Description: The Homogeneous Caspase-6 Assay Kit is a complete assay system designed to measure Caspase-6 activity for screening and profiling applications. It comes in a convenient 96-well format, with all the reagents necessary for 100 fluorescent Caspase-6 activity measurements. In addition, the kit includes purified Caspase-6 enzyme and a potent Caspase-6 inhibitor, Ac-IETD-CHO, for use as a positive and negative control. Using this kit, only one simple step on a microtiter plate is needed to analyze the Caspase-6 activity level. The fluorogenic substrate, Ac-VEID-AFC, is incubated with purified Caspase-6 and the enzymatic activity releases AFC fluorophore that can then be measured using a fluorescence reader.
